Talquetamab plus daratumumab with or without pomalidomide improves PFS in r/r multiple myeloma

Dr Peter Voorhees speaks to ecancer about results from the phase 3 MonumenTAL 3 trial evaluating talquetamab, a GPRC5D targeting bispecific antibody, in combination with daratumumab with or without pomalidomide in patients with relapsed or refractory multiple myeloma after at least one prior line of therapy.

The study explores whether these novel combinations can improve outcomes compared with the standard daratumumab pomalidomide dexamethasone regimen.

At interim analysis, both talquetamab based regimens demonstrated a marked improvement in progression free survival, with sustained benefit across key patient subgroups.

Response rates were higher, with a substantial increase in deep responses including complete response and minimal residual disease negativity, highlighting the potency of these combinations in earlier relapse settings.

Dr Voorhees says that overall survival outcomes also favoured the talquetamab arms, reinforcing the clinical impact of these findings.
