Prof Meletios Dimopoulos speaks to ecancer about results from the phase 3 SUCCESSOR 2 trial evaluating mezigdomide, a novel oral CELMoD, in combination with carfilzomib and dexamethasone in patients with relapsed or refractory multiple myeloma previously exposed to anti CD38 antibodies and lenalidomide.
The triplet regimen significantly improved progression free survival compared with standard carfilzomib and dexamethasone, nearly doubling median PFS and demonstrating consistent benefit across high risk subgroups, including heavily pretreated and refractory patients.
Response rates were substantially higher with the addition of mezigdomide, with deeper responses and increased complete response rates.
While higher rates of haematologic toxicity and infections were observed, the safety profile was manageable in this high risk population.
These findings position mezigdomide based combinations as a promising new standard of care option for patients with limited treatment choices in early relapse settings.
